Flow Cytometry

The Clinical Flow Cytometry Laboratory, headed by Jane Yuan, MD, provides a full complement of immunophenotyping studies for evaluation of hematological malignancies, myelodysplasic syndromes and chronic myeloproliferative disorders, immunodeficiency states, stem cell transplantation, and DNA cell cycle analysis using state-of-the-art multi-parameter and multi-color analysis. A broad menu of testing panels is available for clinical use, and the laboratory can also establish protocols to support clinical and translational research activities. The Clinical Flow Cytometry Laboratory is staffed by medical technologists who are certified by the American Society of Clinical Pathology (ASCP), including two who have earned special qualification in flow cytometry, and by board-certified pathologists with extensive experience in the diagnosis and classification of hematological and immunodeficiency disorders.

The flow cytometry laboratory uses Canto II 8-color instruments from Becton Dickinson and Kaluza Analysis Software from Beckman Coulter for leukemia and lymphoma studies. The approach utilizes Boolean logic to define normal cell types and stages of differentiation based on surface antigen expression patterns. The analysis protocols were built for each antibody cocktail based on the antibodies included. An example of normal bone marrow precursor differentiation is shown above. This approach is especially helpful in identification of myelodysplasia and chronic myeloproliferative disorders.
